John “Hombre” Russell (Paul Newman) is a white man who was brought up by Indians when his parents were killed. He currently lives with them when he learns that he has been bequeathed a small city house. He goes to the city and sells the house for a herd of cattle and wishes to travel to another town by stagecoach. His stagecoach passage is replaced by a train ticket at the last minute, so Hombre sets out to find a new life. During the train ride, bandits come aboard and rob the passengers. They then blow up the train and leave the remaining survivors for dead. Hombre decides to lead the survivors in a quest to find the bandits and recover their valuables.
